Blocking peptide for use with Human Sphingosine Kinase 2, Long Form antibodies.
Incubate antibody neat with at least a 50 fold stoichiometric excess of blocking peptide at 37 Deg. C for 20 minutes (molecular weights of peptide and antibody are ~2.5 kDa and ~160 kDa, respectively). Antibody can then be diluted to a concentration suitable for Western Blot.

Example: 10ul or 10ug of ExalphaÂ’s rabbit anti-human Sphingosine Kinase 2 , Long form is added to 10ug of blocking peptide for a total volume of 20 ul. The mixture is allowed to incubate for 20 minutes at 37 Deg. C prior to dilution in suitable buffer (for Western Blot, etc.).
Preparation and Storage
Product should be stored at -20 degree C. Aliquot to avoid freeze/thaw cycles
Products are stable for one year from purchase when stored properly.
Other Notes
Small volumes of SPHK2 peptide vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Rat sphingosine kinase 1 catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (spp)(sphingosine + ATP = sphingosine 1-phosphate + ADP), a lipid mediator with both intra- and extracellular functions. also acts on d-erythro-sphingosine and to a lesser extent sphinganine, but not other lipids, such as d,l-threo-dihydrosphingosine, n,n-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. The long form of sphingosine kinase 1, an N-terminal extension, is responsible for FceRI-mediated calcium mobilization and may play a role in other ligand mediated signaling events.

This gene encodes one of two sphingosine kinase isozymes that catalyze the phosphorylation of sphingosine into sphingosine 1-phosphate. Sphingosine 1-phosphate mediates many cellular processes including migration, proliferation and apoptosis, and also plays a role in several types of cancer by promoting angiogenesis and tumorigenesis. The encoded protein may play a role in breast cancer proliferation and chemoresistance. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.
