MBS911505 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the spectrin, alpha, non-erythrocytic 1 (alpha-fodrin) (SPTAN1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SPTAN1. The ELISA analytical biochemical technique of the MBS911505 kit is based on SPTAN1 antibody-SPTAN1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SPTAN1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SPTAN1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SPTAN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPTAN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPTAN1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPTAN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Spectrins are a family of filamentous cytoskeletal proteins that function as essential scaffold proteins that stabilize the plasma membrane and organize intracellular organelles. Spectrins are composed of alpha and beta dimers that associate to form tetramers linked in a head-to-head arrangement. This gene encodes an alpha spectrin that is specifically expressed in nonerythrocytic cells. The encoded protein has been implicated in other cellular functions including DNA repair and cell cycle regulation. Mutations in this gene are the cause of early infantile epileptic encephalopathy-5. Alternate splicing results in multiple transcript variants.[provided by RefSeq, Sep 2010]
