Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of Stat3 . No significant cross-reactivity or interference between Stat3 and analogues was observed. Note: Limited by current skills and knowledge, it is difficult for us to complete the crossreactivity detection between Stat3 and all the analogues, therefore, cross reaction may still exist.
Samples
Serum, plasma, tissue homogenates and other biological fluids
Assay Type
Sandwich
Detection Range
0.156-10ng/ml
Sensitivity
<0.094ng/ml
Intra-assay Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Stat3 were tested 20 times on one plate, respectively. Intra-Assay: CV<8%
Inter-assay Precision
Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Stat3 were tested on 3 different plates, 8 replicates in each plate. CV (%) = SD/meanX100. Inter-Assay: CV<10%

Manufactured in an ISO 9001:2008 Certified Laboratory.
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of Stat3 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS760293 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Signal transducer and activator of transcription 3 (Stat3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Stat3. The ELISA analytical biochemical technique of the MBS760293 kit is based on Stat3 antibody-Stat3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Stat3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Stat3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- Stat3 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- Stat3 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRPStreptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Stat3 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Stat3 can be calculated.

UniProt Comments for Stat3
STAT3: transcription factor of the STAT family. Phosphorylated and activated by receptor-associated kinases downstream of many cytokines and growth-factor receptors. Constitutively active in a number of human tumors. Forms homo- or heterodimers that translocate into the nucleus where they regulate transcription. Two alternatively spliced isoforms have been described.
