The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SYVN1. The ELISA analytical biochemical technique of the MBS939550 kit is based on SYVN1 antibody-SYVN1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SYVN1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SYVN1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SYVN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SYVN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SYVN1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SYVN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a protein involved in endoplasmic reticulum (ER)-associated degradation. The encoded protein removes unfolded proteins, accumulated during ER stress, by retrograde transport to the cytosol from the ER. This protein also uses the ubiquitin-proteasome system for additional degradation of unfolded proteins. Sequence analysis identified two transcript variants that encode different isoforms. SYVN1: Acts as an E3 ubiquitin-protein ligase which accepts ubiquitin specifically from endoplasmic reticulum-associated UBC7 E2 ligase and transfers it to substrates, promoting their degradation. Component of the endoplasmic reticulum quality control (ERQC) system also called ER-associated degradation (ERAD) involved in ubiquitin-dependent degradation of misfolded endoplasmic reticulum proteins. Also promotes the degradation of normal but naturally short-lived proteins such as SGK. Protects cells from ER stress-induced apoptosis. Protects neurons from apoptosis induced by polyglutamine-expanded huntingtin (HTT) or unfolded GPR37 by promoting their degradation.
