The ELISA analytical biochemical technique of the MBS903328 kit is based on TAS2R38 antibody-TAS2R38 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TAS2R38 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TAS2R38. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TAS2R38 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TAS2R38 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TAS2R38 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TAS2R38 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a seven-transmembrane G protein-coupled receptor that controls the ability to taste glucosinolates, a family of bitter-tasting compounds found in plants of the Brassica sp. Synthetic compounds phenylthiocarbamide (PTC) and 6-n-propylthiouracil (PROP) have been identified as ligands for this receptor and have been used to test the genetic diversity of this gene. Although several allelic forms of this gene have been identified worldwide, there are two predominant common forms (taster and non-taster) found outside of Africa. These alleles differ at three nucleotide positions resulting in amino acid changes in the protein (A49P, A262V, and V296I) with the amino acid combination PAV identifying the taster variant (and AVI identifying the non-taster variant).
