The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TERT. The ELISA analytical biochemical technique of the MBS943198 kit is based on TERT antibody-TERT antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TERT antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TERT. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TERT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TERT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TERT is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TERT bound in the initial step. The color development is stopped and the intensity of the color is measured.

Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG. The enzyme consists of a protein component with reverse transcriptase activity, encoded by this gene, and an RNA component which serves as a template for the telomere repeat. Telomerase expression plays a role in cellular senescence, as it is normally repressed in postnatal somatic cells resulting in progressive shortening of telomeres. Deregulation of telomerase expression in somatic cells may be involved in oncogenesis. Studies in mouse suggest that telomerase also participates in chromosomal repair, since de novo synthesis of telomere repeats may occur at double-stranded breaks. Alternatively spliced variants encoding different isoforms of telomerase reverse transcriptase have been identified; the full-length sequence of some variants has not been determined.
