MBS910833 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the tissue factor (TF) /coagulation factor VII (F7) complex, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TF. The ELISA analytical biochemical technique of the MBS910833 kit is based on TF antibody-TF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TF/F7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TF/F7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TF/F7 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TF/F7 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes coagulation factor III which is a cell surface glycoprotein. This factor enables cells to initiate the blood coagulation cascades, and it functions as the high-affinity receptor for the coagulation factor VII. The resulting complex provides a catalytic event that is responsible for initiation of the coagulation protease cascades by specific limited proteolysis. Unlike the other cofactors of these protease cascades, which circulate as nonfunctional precursors, this factor is a potent initiator that is fully functional when expressed on cell surfaces. There are 3 distinct domains of this factor: extracellular, transmembrane, and cytoplasmic. This protein is the only one in the coagulation pathway for which a congenital deficiency has not been described. Alternate splicing results in multiple transcript variants.[provided by RefSeq, May 2010]
