The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TLR6. The ELISA analytical biochemical technique of the MBS705756 kit is based on TLR6 antibody-TLR6 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TLR6 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TLR6. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TLR-6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TLR-6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TLR-6 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TLR-6 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor functionally interacts with toll-like receptor 2 to mediate cellular response to bacterial lipoproteins. A Ser249Pro polymorphism in the extracellular domain of the encoded protein may be associated with an increased of asthma is some populations.[provided by RefSeq, Jan 2011]
