The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TMEM195. The ELISA analytical biochemical technique of the MBS7244380 kit is based on TMEM195 antibody-TMEM195 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TMEM195 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TMEM195. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This TMEM195 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse TMEM195. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: TMEM195 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TMEM195 antibody and an TMEM195-HRP conjugate. The assay sample and buffer are incubated together with TMEM195-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TMEM195 concentration since TMEM195 from samples and TMEM195-HRP conjugate compete for the anti-TMEM195 antibody binding site. Since the number of sites is limited, as more sites are occupied by TMEM195 from the sample, fewer sites are left to bind TMEM195-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TMEM195 concentration in each sample is interpolated from this standard curve.
