Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of TPO elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS020824 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Thrombopoietin (TPO) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TPO. The ELISA analytical biochemical technique of the MBS020824 kit is based on TPO antibody-TPO antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TPO antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TPO. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic or diagnostic applications! It is intended to be determinated TPO concentrations in Rat serum, plasma and other body fluids. Using Purified Rat TPO antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add TPO and TPO antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of TPO in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for TPO
Megakaryocytopoiesis is the cellular development process that leads to platelet production. The protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. Mutations in this gene are the cause of thrombocythemia 1. Alternate splicing results in multiple transcript variants of this gene.[provided by RefSeq, Jun 2012]
UniProt Comments for TPO
THPO: Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets. Defects in THPO are the cause of thrombocythemia type 1 (THCYT1). A myeloproliferative disorder characterized by elevated platelet levels due to sustained proliferation of megakaryocytes, and frequently lead to thrombotic and haemorrhagic complications. Belongs to the EPO/TPO family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell cycle regulation; Secreted; Secreted, signal peptide; Cytokine

Chromosomal Location of Human Ortholog: 3q27

Cellular Component: extracellular space

Molecular Function: growth factor activity; hormone activity; cytokine activity

Biological Process: positive regulation of protein kinase B signaling cascade; platelet activation; cell proliferation; multicellular organismal development; myeloid cell differentiation; positive regulation of megakaryocyte differentiation; positive regulation of protein amino acid phosphorylation; blood coagulation

Disease: Thrombocythemia 1
