MBS706000 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the TNF receptor-associated protein 1 (TRAP1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TRAP1. The ELISA analytical biochemical technique of the MBS706000 kit is based on TRAP1 antibody-TRAP1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TRAP1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TRAP1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). TRAP1 - heat shock protein 75 - is a mitochondrial localized homolog of HSP 90. Publications have shown this protein interacts with tumor necrosis factor like receptor and the transcription factor Rb activating inflammatory pathways. Quantitation of cell samples or large sample arrays are made simple with a TRAP 1 ELISA assay. Available in pre-coated multi wells (48) or micrometer plate kits, these ELISA kits show excellent sensitivity and dynamic range for detection of human HSP 75 kDa. All required reagents are provided in this product, including primary antibody, secondary antibodies, wash products and proteins for standard curve generation. Because of high homology within the TNF receptor factor family, such as TNF receptor protein, soluble TNF receptor and TNF receptor II, any assay kit for mitochondrial TRAP1 requires the high level of specificity and lack of cross reaction available in this TRAP1 ELISA kit.

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TRAP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TRAP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TRAP1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TRAP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
