Species Reactivity
This assay has high sensitivity and excellent specificity for detection of UCP2. No significant cross-reactivity or interference between UCP2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between UCP2 and all the analogues, therefore, cross reaction may still exist in some cases.

Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of UCP2 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS7234483 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Mitochondrial uncoupling protein 2 (UCP2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing UCP2. The ELISA analytical biochemical technique of the MBS7234483 kit is based on UCP2 antibody-UCP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect UCP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, UCP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This UCP2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse UCP2.

Principle of the Assay: UCP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-UCP2 antibody and an UCP2-HRP conjugate. The assay sample and buffer are incubated together with UCP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UCP2 concentration since UCP2 from samples and UCP2-HRP conjugate compete for the anti-UCP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by UCP2 from the sample, fewer sites are left to bind UCP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UCP2 concentration in each sample is interpolated from this standard curve.

NCBI Summary for UCP2
Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). UCPs separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. Tissue specificity occurs for the different UCPs and the exact methods of how UCPs transfer H+/OH- are not known. UCPs contain the three homologous protein domains of MACPs. This gene is expressed in many tissues, with the greatest expression in skeletal muscle. It is thought to play a role in nonshivering thermogenesis, obesity and diabetes. Chromosomal order is 5'-UCP3-UCP2-3'. [provided by RefSeq, Jul 2008]
UniProt Comments for UCP2
UCP2: UCP are mitochondrial transporter proteins that create proton leaks across the inner mitochondrial membrane, thus uncoupling oxidative phosphorylation from ATP synthesis. As a result, energy is dissipated in the form of heat. Belongs to the mitochondrial carrier family.
