The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing UCP2. The ELISA analytical biochemical technique of the MBS7234483 kit is based on UCP2 antibody-UCP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect UCP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, UCP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This UCP2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse UCP2.

Principle of the Assay: UCP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-UCP2 antibody and an UCP2-HRP conjugate. The assay sample and buffer are incubated together with UCP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UCP2 concentration since UCP2 from samples and UCP2-HRP conjugate compete for the anti-UCP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by UCP2 from the sample, fewer sites are left to bind UCP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UCP2 concentration in each sample is interpolated from this standard curve.

Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). UCPs separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. Tissue specificity occurs for the different UCPs and the exact methods of how UCPs transfer H+/OH- are not known. UCPs contain the three homologous protein domains of MACPs. This gene is expressed in many tissues, with the greatest expression in skeletal muscle. It is thought to play a role in nonshivering thermogenesis, obesity and diabetes. Chromosomal order is 5'-UCP3-UCP2-3'. [provided by RefSeq, Jul 2008]
