Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ULBP2. The ELISA analytical biochemical technique of the MBS734130 kit is based on ULBP2 antibody-ULBP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ULBP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ULBP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: ULBP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ULBP2 antibody and an ULBP2-HRP conjugate. The assay sample and buffer are incubated together with ULBP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ULBP2 concentration since ULBP2 from samples and ULBP2-HRP conjugate compete for the anti-ULBP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ULBP2 from the sample, fewer sites are left to bind ULBP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ULBP2 concentration in each sample is interpolated from this standard curve.

This gene encodes a major histocompatibility complex (MHC) class I-related molecule that binds to the NKG2D receptor on natural killer (NK) cells to trigger release of multiple cytokines and chemokines that in turn contribute to the recruitment and activation of NK cells. The encoded protein undergoes further processing to generate the mature protein that is either anchored to membrane via a glycosylphosphatidylinositol moiety, or secreted. Many malignant cells secrete the encoded protein to evade immunosurveillance by NK cells. This gene is located in a cluster of multiple MHC class I-related genes on chromosome 6. ULBP2: Ligand for the NKG2D receptor, together with at least ULBP1 and ULBP3. ULBPs activate multiple signaling pathways in primary NK cells, resulting in the production of cytokines and chemokines. Binding of ULBPs ligands to NKG2D induces calcium mobilization and activation of the JAK2, STAT5, ERK and PI3K kinase/Akt signal transduction pathway. In CMV infected cells, interacts with soluble CMV glycoprotein UL16. The interaction with UL16 blocked the interaction with the NKG2D receptor, providing a mechanism by which CMV infected cells might escape the immune system. UL16 also causes ULBP2 to be retained in the ER and cis- Golgi apparatus so that it does not reach the cell surface. Belongs to the MHC class I family.
