Intended Uses: This VPS-29 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human VPS-29. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: VPS-29 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VPS-29 antibody and an VPS-29-HRP conjugate. The assay sample and buffer are incubated together with VPS-29-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VPS-29 concentration since VPS-29 from samples and VPS-29-HRP conjugate compete for the anti-VPS-29 antibody binding site. Since the number of sites is limited, as more sites are occupied by VPS-29 from the sample, fewer sites are left to bind VPS-29-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VPS-29 concentration in each sample is interpolated from this standard curve.

This gene belongs to a group of vacuolar protein sorting (VPS) genes that, when functionally impaired, disrupt the efficient delivery of vacuolar hydrolases. The protein encoded by this gene is a component of a large multimeric complex, termed the retromer complex, which is involved in retrograde transport of proteins from endosomes to the trans-Golgi network. This VPS protein may be involved in the formation of the inner shell of the retromer coat for retrograde vesicles leaving the prevacuolar compartment. Alternative splice variants encoding different isoforms and representing non-protein coding transcripts have been found for this gene. [provided by RefSeq, Aug 2013]
