The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing WNT11. The ELISA analytical biochemical technique of the MBS281148 kit is based on WNT11 antibody-WNT11 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect WNT11 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, WNT11. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: For the quantitative detection of Human Protein Wnt-11 (WNT11) concentration in serum, plasma, cell culture supernates and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate WNT11 in Human serum, plasma, cell culture supernates. An antibody specific for WNT11 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WNT11 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WNT11 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT11 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 97%, 85%, and 63% amino acid identity with mouse, chicken, and Xenopus Wnt11 protein, respectively.
