The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing WNT4. The ELISA analytical biochemical technique of the MBS934091 kit is based on WNT4 antibody-WNT4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect WNT4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, WNT4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for WNT4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WNT4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WNT4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family, and is the first signaling molecule shown to influence the sex-determination cascade. It encodes a protein which shows 98% amino acid identity to the Wnt4 protein of mouse and rat. This gene and a nuclear receptor known to antagonize the testis-determining factor play a concerted role in both the control of female development and the prevention of testes formation. This gene and another two family members, WNT2 and WNT7B, may be associated with abnormal proliferation in breast tissue. Mutations in this gene can result in Rokitansky-Kuster-Hauser syndrome and in SERKAL syndrome. [provided by RefSeq, Jul 2008]

May be a signaling molecule which affects the development of discrete regions of tissues. Is likely to signal over only few cell diameters. Overexpression may be associated with abnormal proliferation in human breast tissue. Defects in WNT4 are a cause of Rokitansky-Kuster-Hauser syndrome (RKH syndrome); also called Mayer- Rokitansky-Kuster-Hauser syndrome (MRKH syndrome or MRKH anomaly). RKH syndrome is characterized by utero-vaginal atresia in otherwise phenotypically normal female with a normal 46,XX karyotype.
