The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing WNT9A. The ELISA analytical biochemical technique of the MBS942617 kit is based on WNT9A antibody-WNT9A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect WNT9A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, WNT9A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for WNT9A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WNT9A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WNT9A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT9A bound in the initial step. The color development is stopped and the intensity of the color is measured.

The WNT gene family consists of structurally related genes that encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It is expressed in gastric cancer cell lines. The protein encoded by this gene shows 75% amino acid identity to chicken Wnt14, which has been shown to play a central role in initiating synovial joint formation in the chick limb. This gene is clustered with another family member, WNT3A, in the chromosome 1q42 region. [provided by RefSeq, Jul 2008]
