The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing XDH. The ELISA analytical biochemical technique of the MBS918183 kit is based on XDH antibody-XDH antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect XDH antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, XDH. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay||This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for XDH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any XDH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XDH is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XDH bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the xanthine dehydrogenase protein family. The encoded protein has been identified as a moonlighting protein based on its ability to perform mechanistically distinct functions. The encoded protein exists as two distinct enzymatic forms, either as xanthine dehydrogenase, or as xanthine oxidase, and functions in purine degradation. Additional studies also suggest a role in adipogenesis, and a function as a structural protein in milk fat droplets in the lactating mammary gland. [provided by RefSeq, Jan 2014]

XDH: Key enzyme in purine degradation. Catalyzes the oxidation of hypoxanthine to xanthine. Catalyzes the oxidation of xanthine to uric acid. Contributes to the generation of reactive oxygen species. Has also low oxidase activity towards aldehydes (in vitro). Defects in XDH are the cause of xanthinuria type 1 (XU1). Xanthinuria is characterized by excretion of very large amounts of xanthine in the urine and a tendency to form xanthine stones. Uric acid is strikingly diminished in serum and urine. XU1 is due to isolated xanthine dehydrogenase. XU1 patients can metabolize allopurinol. Belongs to the xanthine dehydrogenase family.
