Species Reactivity
Specificity
This assay has high sensitivity and excellent specificity for detection of YWHAG. No significant cross-reactivity or interference between YWHAG and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between YWHAG and all the analogues, therefore, cross reaction may still exist in some cases.

Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of YWHAG elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS7238731 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the 14 3 3 protein gamma (YWHAG) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing YWHAG. The ELISA analytical biochemical technique of the MBS7238731 kit is based on YWHAG antibody-YWHAG antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect YWHAG antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, YWHAG. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This YWHAG ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human YWHAG. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: YWHAG ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-YWHAG antibody and an YWHAG-HRP conjugate. The assay sample and buffer are incubated together with YWHAG-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the YWHAG concentration since YWHAG from samples and YWHAG-HRP conjugate compete for the anti-YWHAG antibody binding site. Since the number of sites is limited, as more sites are occupied by YWHAG from the sample, fewer sites are left to bind YWHAG-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The YWHAG concentration in each sample is interpolated from this standard curve.

NCBI Summary for YWHAG
This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 100% identical to the rat ortholog. It is induced by growth factors in human vascular smooth muscle cells, and is also highly expressed in skeletal and heart muscles, suggesting an important role for this protein in muscle tissue. It has been shown to interact with RAF1 and protein kinase C, proteins involved in various signal transduction pathways. [provided by RefSeq, Jul 2008]
UniProt Comments for YWHAG
14-3-3 gamma: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. Homodimer. Interacts with SAMSN1. Interacts with RAF1, SSH1 and CRTC2/TORC2. Interacts with ABL1 (phosphorylated form); the interaction retains it in the cytoplasm. Interacts with GAB2. Interacts with MDM4 (phosphorylated); negatively regulates MDM4 activity toward TP53. Interacts with PKA-phosphorylated AANAT. Highly expressed in brain, skeletal muscle, and heart. Belongs to the 14-3-3 family.
