ELISA KitBRG1-associated factor (BAF) 155, one member of SWI/SNF complexes, is found in cyclin E complexes and is phosphorylated by cyclin E-associated kinase. The BAF155 and BAF170 proteins are highly homologous and exist either as homodimers (BAF155/155 or BAF170/170) or as heterodimers (BAF155/170) through a leucine zipper motif in the cell. BAF155 contains two highly conserved motifs that are found in chromatin associated proteins. SWIRM (Swi3, Rsc8, and Moria), one domain of BAF155, is believed to adopt an _-helical structure and mediate specific protein-protein interaction. The other motif, termed the SANT (Swi3, Ads2, N-coR, and TFIIIB) domain, is believed to function as a histone tail binding module. Since the BAF155 is located on chromosome 3p221.31, which contains some suspected tumor suppressor genes such as FUS1 and SEM3B, the loss of BAF155 might contribute to tumor development.
A study found that an epigenetic enzyme coactivator associated arginine methyltransferase 1 (CARM1) is highly expressed in triple-negative breast cancers (TNBC) and its high expression correlates with poor prognosis. Subsequently the study identified BRG1-Associated Factor 155 (BAF155) as a novel CARM1 substrate. Transcriptome analyses of BAF155 methylation competent and defective cell lines identified many genes involved in regulating cancer metastasis are methyl-BAF155 targets. A study mapped global me-BAF155 genomic association sites using ChIP-seq and discovered that me-BAF155 was highly enriched in super-enhancers (SEs), regulatory elements on chromatin that are known to drive high transcription of many oncogenes. The data of the study suggest that me-BAF155 is an important epigenetic target for treatment of cancer metastasis in TNBC cells.
