Other Notes

Small volumes of tPA active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Searchable Terms for tPA purchase

Human tissue plasminogen activator (tPA) is also known as alteplase, TPA, T-PA, t-PA, PLAT and tissue-type plasminogen activator.

tPA is secreted serine protease (EC 3.4.21.68), naturally occurring in endothelial cells of blood vessels; it converts proenzyme plasminogen to its active form, plasmin, enzyme responsible for breakdown of blood clots ; increased activity of tPA may cause hyperfibrinolysis and excessive bleeding including intracranial haemorrhage, and reduced activity may result in thrombosis or embolism due to hypofibrinolysis.

tPA is recognized for its role in thrombolysis (blood clots breakdown) and administered at ischemic (ischaemic) conditions such as acute ischemic (ischaemic) stroke. Due to tPA specific functions in blood clotting system, it’s used clinically to treat thrombolytic or embolic stroke, acute myocardial infarction, pulmonary embolism, and its use is contradicted in cases when bleeding is expected such as head trauma or hemorrhagic stroke.

tPA is recombinant protein produced in CHO cells as single-chain glycosylated polypeptide containing 527 amino acids and with molecular mass of 59kDA, and is purified by proprietary chromatographic techniques; its purity is greater than 98% (by RP-HPLC and SDS-PAGE) and activity is 580,000IU/mg.

It’s provided as lyophilized preparation and is stable for up to 3 weeks at ambient temperature; recommended storage is below -18°C, dessicated.

NCBI Summary for tPA

This gene encodes tissue-type plasminogen activator, a secreted serine protease which converts the proenzyme plasminogen to plasmin, a fibrinolytic enzyme. Tissue-type plasminogen activator is synthesized as a single chain which is cleaved by plasmin to a two chain disulfide linked protein. This enzyme plays a role in cell migration and tissue remodeling. Increased enzymatic activity causes hyperfibrinolysis, which manifests as excessive bleeding; decreased activity leads to hypofibrinolysis which can result in thrombosis or embolism. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

UniProt Comments for tPA

PLAT: Converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling and degradation, in cell migration and many other physiopathological events. Plays a direct role in facilitating neuronal migration. Increased activity of TPA results in increased fibrinolysis of fibrin blood clots that is associated with excessive bleeding. Defective release of TPA results in hypofibrinolysis that can lead to thrombosis or embolism. Belongs to the peptidase S1 family. 4 isoforms of the human protein are produced by alternative splicing.
