Immunohistochemistry (IHC) of anti-AGO2 antibody

IHC image of MBS1493111 diluted at 1:1200 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Chromatin Immunoprecipitation (ChIP) of anti-AGO2 antibody

Chromatin Immunoprecipitation Hela(1.1^106)were cross-linked with formaldehyde, sonicated, and immunoprecipitated with 4ug anti-AgO2 or a control normal rabbit IgG . The resulting ChIP DNA was quantified using real-time PCR with primers against the SLC1A5 promoter.

NCBI Summary for AGO2

This gene encodes a member of the Argonaute family of proteins which play a role in RNA interference. The encoded protein is highly basic, and contains a PAZ domain and a PIWI domain. It may interact with dicer1 and play a role in short-interfering-RNA-mediated gene silencing. Multiple transcript variants encoding different isoforms have been found for this gene. UniProt Comments for AGO2

AGO2: the endonuclease in RNA-induced silencing complexes (RISC) that cleaves siRNA/mRNA heteroduplexes bound to RISC. Ago2, along with Dicer and TRBP, are major components of RISC. Interacts with Dicer1 through its Piwi domain. Required for proper fibroblast growth factor signaling during gastrulation. Essential for embryonic development as well as RNA-mediated gene silencing (RNAi).
