KIM1 (HAVCR1) ELISA Kit for Mouse

Principle of the Assay: Picokine™ Mouse Havcr1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Havcr1 with a 96-well strip plate that is pre-coated with antibody specific for Havcr1. The detection antibody is a biotinylated antibody specific for Havcr1. The capture antibody is polyclonal antibody from goat and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Mouse Havcr1 with immunogen: Expression system for standard: NS0; Immunogen sequence: Y22-T212. The kit is analytically validated with ready-to-use reagents. To measure Mouse Havcr1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Havcr1 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Havcr1 in the sample.

Background/Introduction: KIM1 (TIM-1), also known as Hepatitis A virus cellular receptor 1, is a protein that in Mouses is encoded by the HAVCR1 gene.[1][2][3] Infection of canine osteogenic sarcoma cells expressing HAVCR1 with HAV led Feigelstock et al. (1998) to conclude that the protein is indeed a receptor for the virus. Immunofluorescence microscopy demonstrated internalization of HAV by dog cells expressing HAVCR1. Using a monoclonal antibody to mouse Tim1, Umetsu et al. (2005) showed that Tim1 was expressed after activation of naive T cells and on T cells differentiated in Th2-polarizing conditions. By homology of synteny with the mouse Tim1 gene and database analysis, McIntire et al. (2001) mapped the HAVCR1 gene to 5q33.2. The standard used in this kit is recombinant mouse KIM-1, constituting from Y22-T212 amino acids, and 191amino acids with the molecular mass of 21.8KDa.
