Principle of the Assay: The Picokine Human IL17F Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human IL17F with a 96-well strip plate that is pre-coated with antibody specific for IL17F. The detection antibody is a biotinylated antibody specific for IL17F. The capture antibody is A monoclonal antibody from mouse, the detection antibody is a biotinylated detection polyclonal antibody from goat. The kit contains recombinant Human IL17F with immunogen: Expression system for standard: NSO, Immunogen sequence: R31-Q163. The kit is analytically validated with ready to use reagents. To measure Human IL17F, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human IL17F in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human IL17F in the sample!!Background/Introduction: Members of the interleukin-17 family, such as IL17F, are involved in the regulation of normal versus aberrant T-cell responses.. It is mapped to 6p12.2. The protein encoded by this gene is a cytokine that shares sequence similarity with IL17. This cytokine is expressed by activated T cells, and has been shown to stimulate the production of several other cytokines, including IL6, IL8, and CSF2/GM_CSF. This cytokine is also found to inhibit the angiogenesis of endothelial cells and induce endothelial cells to produce IL2, TGFB1/TGFB, and monocyte chemoattractant protein-1.
