Intended Uses
This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic applications! It is intended to be determinated CASPASE-2 concentrations in Bovine serum, plasma and other body fluids. Using Purified Bovine CASPASE-2 antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add CASPASE-2 and CASPASE-2 antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of CASPASE-2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Other Notes

Small volumes of CASPASE-2 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS017723 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Caspase 2 (CASPASE-2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CASPASE-2. The ELISA analytical biochemical technique of the MBS017723 kit is based on CASPASE-2 antibody-CASPASE-2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CASPASE-2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CASPASE-2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for CASPASE-2

This gene encodes the evolutionarily ancient and most conserved member of the cysteine proteases that plays important role in stress-induced apoptosis, DNA repair and tumor suppression. Mice lacking the encoded protein develop normally but display cell type-specific apoptotic defects. Germ cells and oocytes from such mice were found to be resistant to cell death after treatment with chemotherapeutic drugs. [provided by RefSeq, Apr 2015]

UniProt Comments for CASPASE-2

CASP2: Involved in the activation cascade of caspases responsible for apoptosis execution. Might function by either activating some proteins required for cell death or inactivating proteins necessary for cell survival. Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a p18 subunit and a p12 subunit. Interacts with LRDD. Expressed at higher levels in the embryonic lung, liver and kidney than in the heart and brain. In adults, higher level expression is seen in the placenta, lung, kidney, and pancreas than in the heart, brain, liver and skeletal muscle. Belongs to the peptidase C14A family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Protease; Apoptosis; EC 3.4.22.55

Cellular Component: membrane; mitochondrion; cytoplasm; intracellular; nucleus

Molecular Function: peptidase activity; protein domain specific binding; protein binding; hydrolase activity; cysteine-type endopeptidase activity; cysteine-type peptidase activity

Biological Process: caspase activation; regulation of apoptosis; DNA damage response, signal transduction resulting in induction of apoptosis; apoptosis; positive regulation of apoptosis; positive regulation of neuron apoptosis; germ cell programmed cell death; protein processing; proteolysis; DNA damage response, signal transduction by p53 class mediator resulting in cell cycle arrest; negative regulation of apoptosis
