Other Notes

Small volumes of GZMS-A elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS017784 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Granzymes A (GZMS-A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GZMS-A. The ELISA analytical biochemical technique of the MBS017784 kit is based on GZMS-A antibody-GZMS-A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GZMS-A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GZMS-A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for GZMS-A

Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein described here is a T cell- and natural killer cell-specific serine protease that may function as a common component necessary for lysis of target cells by cytotoxic T lymphocytes and natural killer cells. UniProt Comments for GZMS-A

GZMA: This enzyme is necessary for target cell lysis in cell- mediated immune responses. It cleaves after Lys or Arg. Cleaves APEX1 after 'Lys-31' and destroys its oxidative repair activity. Involved in apoptosis. Homodimer; disulfide-linked. Interacts with APEX1. Dexamethasone (DEX) induces expression of isoform beta and represses expression of isoform alpha. The alteration in expression is mediated by binding of glucocorticoid receptor to independent promoters adjacent to the alternative first exons of isoform alpha and isoform beta. Belongs to the peptidase S1 family. Granzyme subfamily. 2 isoforms of the human protein are produced by alternative promoter.

Protein type: EC 3.4.21.78; Secreted; Secreted, signal peptide; Protease; Apoptosis

Chromosomal Location of Human Ortholog: 5q11-q12

Cellular Component: extracellular region; immunological synapse; nucleus

Molecular Function: protein binding; protein homodimerization activity; serine-type endopeptidase activity

Biological Process: proteolysis involved in cellular protein catabolic process; positive regulation of apoptosis; apoptosis; cytolysis; negative regulation of oxidoreductase activity; negative regulation of endodeoxyribonuclease activity; immune response; negative regulation of DNA binding
