Western Blot (WB) of anti-BIK antibody

Figure 1. Western blot analysis of Bik using anti-Bik antibody (MBS178226).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: Rat Spleen Tissue Lysate,
Lane 2: Rat Gaster Tissue Lysate,
Lane 3: Rat Intestine Tissue Lysate,
Lane 4: MCF-7 Whole Cell Lysate,
Lane 5: A549 Whole Cell Lysate,
Lane 6: SKOV Whole Cell Lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Bik antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Bik at approximately 22KD. The expected band size for Bik is at 22KD.

Other Notes

Small volumes of anti-BIK antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for BIK

The protein encoded by this gene shares a critical BH3 domain with other death-promoting proteins, such as BID, BAK, BAD and BAX, that is required for its pro-apoptotic activity, and for interaction with anti-apoptotic members of the BCL2 family, and viral survival-promoting proteins. Since the activity of this protein is suppressed in the presence of survival-promoting proteins, it is suggested as a likely target for anti-apoptotic proteins. [provided by RefSeq, Sep 2011]

UniProt Comments for BIK

BIK: accelerates programmed cell death. Binding to the apoptosis repressors Bcl-xL, Bcl-2, or BHRF1 (the Epstein-Barr virus homolog of Bcl-2) suppresses its death-promoting activity. Contains 1 Bcl-2 homology 3 (BH3) domain. Does not interact with Bax.
