Specificity
This product detects an epitope within the internal region of CD14, a 55kD receptor for endotoxin and other bacterial cell wall components. Upon binding of various ligands, CD14 acts as a multiligand pattern recognition receptor, inducing specific coassembly of additional receptors, including MD-2 and TLR4 as part of the lipopolysaccharide (LPS) receptor.

The LPS receptor is expressed strongly on the surface of monocytes and weakly on the surface of granulocytes, and is also expressed by most tissue macrophages. Activation of the LPS receptor leads to NF-kappa-B activation, cytokine secretion and the inflammatory response. CD14 also up-regulates cell surface molecules, including adhesion molecules, and has been shown to bind apoptotic cells.
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Application Notes for anti-CD14 antibody
Western Blot: This item detects a band of approximately 55kD in human lung, lymph node and tonsil lysates.
ELISA: Minimum Dilution: 1/128,000
Immunohistology - Paraffin: Minimum Dilution: 4; Maximum Dilution: 6 ug/ml; Application Note: Heat-mediated antigen retrieval is required prior to staining paraffin sections with this antibody. Citrate buffer, pH6.0, is recommended for this purpose.
Western Blotting: Minimum Dilution: 1; Maximum Dilution: 3 ug/ml

Other Notes

Small volumes of anti-CD14 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CD14

The protein encoded by this gene is a surface antigen that is preferentially expressed on monocytes/macrophages. It cooperates with other proteins to mediate the innate immune response to bacterial lipopolysaccharide. Alternative splicing results in multiple transcript variants encoding the same protein.
