This item detects the p65 (RelA) subunit of nuclear factor kappa-B (NFkB) when phosphorylated at serine 536. NFkB is a heterodimeric DNA binding protein which was originally identified consisting of p65 and p50 (NFkB1) subunits. Research has led to the discovery of other subunits, including p52 (NFkB2), cRel, and RelB.

NFkB is ubiquitously expressed, and functions as a second messenger which upon activation leads to the transcription of a number of genes. NFkB is activated by a number of agents, including pro-inflammatory cytokines and bacterial lipopolysaccharide, and is essential for the maturation of T cells and regulation of their survival and activation.

Phosphorylation of serine 536 contributes to NFkB transactivation and increases p65 transcription activity.
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

ELISA (EIA), Immunohistology Paraffin, Western Blot (WB)*
Application Notes for anti-NFkB p65 antibody
Western Blot: This item detects a band of approximately 65kDa in the appropriate cell lysate.
ELISA: Minimum Dilution: 1/500; Maximum Dilution: 1/3000
Immunohistology - Paraffin: Minimum Dilution: 1/50; Maximum Dilution: 1/250
Western Blotting: Minimum Dilution: 1/100; Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, threonine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

Other Notes

Small volumes of anti-NFkB p65 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for NFkB p65

NF-kappa-B is a ubiquitous transcription factor involved in several biological processes. It is held in the cytoplasm in an inactive state by specific inhibitors. Upon degradation of the inhibitor, NF-kappa-B moves to the nucleus and activates transcription of specific genes. NF-kappa-B is composed of NFKB1 or NFKB2 bound to either REL, RELA, or RELB. The most abundant form of NF-kappa-B is NFKB1 complexed with the product of this gene, RELA. Four transcript variants encoding different isoforms have been found for this gene. UniProt Comments for NFkB p65

NFkB-p65: a subunit of NF-kappa-B transcription complex, which plays a crucial role in inflammatory and immune responses. The inhibitory effect of I-kappa-B upon NF-kappa-B in the cytoplasm is exerted primarily through the interaction with p65. P65 shows a weak DNA-binding site which could contribute directly to DNA binding in the NF-kappa-B complex. There are five NFkB proteins in mammals (RelA/NFkB-p65, RelB, c-Rel, NF-_B1/NFkB-p105, and NF-_B2/NFkB-p100). They form a variety of homodimers and heterodimers, each of which activates its own characteristic set of genes. Three splice-variant isoforms have been identified.
