Product Overview

Product Name
IL-1RA (IL1RN), Polyclonal Antibody
Full Product Name
RABBIT ANTI HUMAN INTERLEUKIN-1RA:Biotin
Product Gene Name
Clonality
Polyclonal
Isotype
IgG
Host
Rabbit
Form/Format
Biotin
Purified IgG conjugated to Biotin - lyophilised
Concentration
IgG concentration 0.1mg/ml after reconstitution (lot specific)
Specificity
This product specifically recognises human Interleukin-1 receptor antagonist (IL-1RA), a naturally occurring inflammatory inhibitor protein, produced by monocytes, macrophages, neutrophils, corneal epithelial cells and fibroblasts, which inhibits the activity of IL-1 alpha and IL-1 beta, by competitively blocking binding to cell surface receptors for IL-1.

Genetic variations in IL-1RA gene (IL1RN) are associated with susceptibility to microvascular complications of diabetes type 4 (MVCD4), and IL-1RA has therapeutic applications in rheumatoid arthritis, asthma, psoriasis, sepsis, chronic myelogenous leukaemia and inflammatory bowel disease.
Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.
Molecular Weight
16,142 Da
OMIM
147679
3D Structure
ModBase 3D Structure for P18510
Reconstitution
Reconstitute with 0.5ml sterile PBS containing 0.1% bovine serum albumin Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended.
Immunogen
Recombinant human IL-1RA
Buffer Solution||Phosphate buffered saline
Antiserum Preparation
Antiserum to human IL-1RA were raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
Target Species
Human
Applications Tested/Suitable for anti-IL1RN antibody
ELISA (EIA), Western Blot (WB)
Application Notes for anti-IL1RN antibody
ELISA: This biotinylated human IL-1RA antibody may be used in a direct ELISA or as the detection reagent in a sandwich ELISA with a purified human IL-1RA antibody as the capture reagent and recombinant human IL-1RA as the standard.
Western Blot: This biotinylated human IL-1RA antibody may be used in Western Blotting applications under either reducing or non-reducing conditions with recombinant human IL-1RA as the positive control.
ELISA: Minimum Dilution: 0.25; Maximum Dilution: 1.0ug/ml
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml

Other Notes

Small volumes of anti-IL1RN antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL1RN

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer. Four alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

UniProt Comments for IL1RN

IL1RN: Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signaling. Has no interleukin-1 like activity. Binds functional interleukin-1 receptor IL1R1 with greater affinity than decoy receptor IL1R2; however, the physiological relevance of the latter association is unsure. The intracellular form of IL1RN is predominantly expressed in epithelial cells. 4 isoforms of the human protein are produced by alternative splicing.
