LEPTIN HUMAN Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS232445

Leptin is a major adipose-derived hormone and member of the leptin family, which plays a key role in appetite control and the regulation of energy expenditure. Secreted leptin associates with proteins within the bloodstream and is transported to the brain,… More details

Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Reconstitution
Reconstitute with 1.0 ml distilled water. Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For extended storage, the addition of 0.1% bovine serum abumin (BSA) is recommended.
Preparation||Purified recombinant leptin expressed in E Coli. Sterile filtered through a 0.2 micron filter.
Buffer Solution
Lyophilized with 0.1% TFA
Activity
This item is biologically active in the ob/ob mouse obesity model. The ob/ob mice were treated via intraperitoneal injection once daily at a dose of 5 ug Leptin/gm of body weight for 7 days. Significant effects on body weight, food consumption, and plasma glucose levels were observed to saline-treated controls.
Target Species
Human

ELISA (EIA), Functional Assays (FN), Western Blot (WB)
Application Notes for LEPTIN active protein
ELISA: Recombinant human leptin may be use as a standard for ELISA applications with either a purified human leptin antibody or a biotinylated human leptin antibody.
Western Blot: Recombinant human leptin may be used as the positive control for Western Blotting applications with either a purified human leptin antibody or a biotinylated human leptin antibody.
ELISA: Minimum Dilution: 0.2; Maximum Dilution: 0.4ng/well
Western Blotting: Minimum Dilution: 1.5; Maximum Dilution: 3.0ng/lane

Other Notes

Small volumes of LEPTIN active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for LEPTIN

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. UniProt Comments for LEPTIN

leptin: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Belongs to the leptin family.
