Immunohistochemistry (IHC) of anti-SPRY4 antibody

Anti-SPRY4 antibody IHC of human heart. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 5 ug/ml.
Rabbit Anti-SPRY4 Polyclonal Antibody – Cat# MBS240219

Western Blot (WB) of anti-SPRY4 antibody

Anti-Sprouty-4 Antibody - Western Blot. Western blot of Affinity Purified anti-Sprouty-4 antibody shows detection of a doublet band ~35 kD corresponding to human Sprouty-4. Approximately 30 ug of HeLa (lane 1), SW13 (lane 2) and C2C12 (Lane 3) whole cell lysates were separated by 10% SDS-PAGE and transferred onto nitrocellulose. After blocking with 5% non-fat dry milk in TBST the membrane was probed with the primary antibody diluted to 1:100 overnight at 4C followed by washes and reaction with a 1:5000 dilution of HRP conjugated Gt-a-Rabbit IgG [H&L] MX for 1 h at room temperature. Signal was procebetaed by ECL (Pierce). Other detection systems will yield similar results.

NCBI Summary for SPRY4

This gene encodes a member of a family of cysteine- and proline-rich proteins. The encoded protein is an inhibitor of the receptor-transduced mitogen-activated protein kinase (MAPK) signaling pathway. Activity of this protein impairs the formation of active GTP-RAS. Nucleotide variation in this gene has been associated with hypogonadotropic hypogonadism 17 with or without anosmia. Alternative splicing results in a multiple transcript variants. UniProt Comments for SPRY4

SPRY4: Suppresses the insulin receptor and EGFR-transduced MAPK signaling pathway, but does not inhibit MAPK activation by a constitutively active mutant Ras. Probably impairs the formation of GTP-Ras. Inhibits Ras-independent, but not Ras-dependent, activation of RAF1. Belongs to the sprouty family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Inhibitor; Adaptor/scaffold

Chromosomal Location of Human Ortholog: 5q31.3

Cellular Component: Golgi apparatus; focal adhesion; cytoplasm

Molecular Function: protein binding

Biological Process: negative regulation of MAP kinase activity; multicellular organismal development

Disease: Hypogonadotropic Hypogonadism 17 With Or Without Anosmia
