Immunohistochemistry (IHC) of anti-PPP1R13L antibody

Anti-PPP1R13L / IASPP antibody IHC of human liver. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody concentration 5 ug/ml.
Rabbit Anti-PPP1R13L Polyclonal Antibody – Cat# MBS240529

Western Blot (WB) of anti-PPP1R13L antibody

Anti-iASPP Antibody - Western Blot. Western blot of affinity purified anti-iASPP antibody shows detection of a band at ~100 kD (arrowhead) corresponding to isoform 1 of iASPP in MCF7 whole cell lysates. Preincubation with immunizing peptide blocks specific band staining (data not shown). Approximately 35 ug of lysate was separated by 4-20% Tris Glycine SDS-PAGE. After blocking, the membrane was probed with the primary antibody diluted to 1:1500 in 5% BLOTTO/PBS overnight at 4C. The membrane was washed and reacted with a 1:10000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] (for 45 min at room temperature (800 nm channel, green). Molecular weight estimation was made by comparison to prestained MW markers. IRDye800 fluorescence image was captured using the Odyssey Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Other Notes

Small volumes of anti-PPP1R13L antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PPP1R13L

IASPP is one of the most evolutionarily conserved inhibitors of p53 (TP53; MIM 191170), whereas ASPP1 (MIM 606455) and ASPP2 (MIM 602143) are activators of p53.[supplied by OMIM, Mar 2008]

UniProt Comments for PPP1R13L

PPP1R13L: an oncoprotein that interacts with and inhibits NF-kappaB, Sp1 and p53. Plays a central role in regulation of apoptosis and transcription. May inhibit p53 by preventing its association with ASPP1 or ASPP2, thereby suppressing the subsequent activation of apoptosis. Required for neuronal survival after axonal injury. Two alternatively spliced isoforms have been described. Isoform 1 is preferrentially found in the cytoplasm; isoform 2 is nuclear.

Protein type: Phosphatase, regulatory subunit; Apoptosis; Transcription, coactivator/corepressor

Chromosomal Location of Human Ortholog: 19q13.32

Cellular Component: intercellular bridge; cytoplasm; cell junction; nucleus

Molecular Function: identical protein binding; protein binding; transcription factor binding; transcription corepressor activity

Biological Process: transcription, DNA-dependent; apoptosis; multicellular organism growth; negative regulation of transcription from RNA polymerase II promoter; multicellular organismal homeostasis; embryonic camera-type eye development; post-embryonic development; cardiac muscle contraction; hair cycle
