Immunogen Description
Mixture of five recombinant constructs containing the entire C-terminal region of human alpha-II spectrin (amino acids 676-2,400).
Immunogen Type
Recombinant protein
Immunogen
SPTAN1 / Alpha Fodrin antibody was raised against mixture of five recombinant constructs containing the entire C-terminal region of human alpha-II spectrin (amino acids 676-2,400). The 9th spectrin repeat also includes a Src-homology 3 domain, and the extreme C-terminus beyond the 20th spectrin repeat contains two EF hand calcium binding motifs. These constructs were expressed in and purified from E Coli.
Applications Tested/Suitable for anti-SPTAN1 antibody
Immunohistochemistry (IHC - Paraffin), Immunocytochemistry (ICC), Immunofluorescence (IF), Western Blot (WB)
Application Notes for anti-SPTAN1 antibody
ICC (1:500 - 1:1000), IF (1:500 - 1:1000), IHC-P (1:100), WB (1:5000 - 1:10000)
Usage: The serum can be diluted to 1:500-1:1000 for immunofluorescence staining and 1:5000-1:10000 for western blotting. On western blots look for a major band at 240kDa, depending on the species.

Immunohistochemistry (IHC) of anti-SPTAN1 antibody

Anti-SPTAN1 / Alpha Fodrin antibody IHC staining of human small intestine, submucosal plexus. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody dilution 1:100.

Immunocytochemistry (ICC) of anti-SPTAN1 antibody

Mixed neuron-glial cultures stained with rabbit antibody to alpha-II spectrin (green) and mouse polyclonal antibody to Nestin (red). The alpha-II spectrin antibody stains numerous axonal and dendritic profiles in these cultures, clearly revealing the submembranous cytoskeleton. Since alpha-II spectrin is specific for neurons in the CNS, the glial cells in this culture are not recognized by this antibody.

Western Blot (WB) of anti-SPTAN1 antibody

Western blots of mouse sciatic nerve extract (lanes 1 and 2) and brain homogenate (3). A prominent band running at 240 kDa represents the intact alpha-II spectrin heavy chain. More minor bands below this likely represent in vivo proteolytic fragments of alpha-II spectrin.

Other Notes

Small volumes of anti-SPTAN1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for SPTAN1

Spectrins are a family of filamentous cytoskeletal proteins that function as essential scaffold proteins that stabilize the plasma membrane and organize intracellular organelles. Spectrins are composed of alpha and beta dimers that associate to form tetramers linked in a head-to-head arrangement. This gene encodes an alpha spectrin that is specifically expressed in nonerythrocytic cells. The encoded protein has been implicated in other cellular functions including DNA repair and cell cycle regulation. Mutations in this gene are the cause of early infantile epileptic encephalopathy-5. Alternate splicing results in multiple transcript variants.[provided by RefSeq, Sep 2010]

UniProt Comments for SPTAN1

SPTAN1: a cytoskeletal protein which seems to be involved in secretion, interacts with calmodulin in a calcium-dependent manner and is thus candidate for the calcium-dependent movement of the cytoskeleton at the membrane. Capable to forming dimers which can further associate to tetramers.
