Immunohistochemistry (IHC) of anti-TSC2 antibody

Anti-Tuberin / TSC2 antibody IHC staining of human liver. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody dilution 1:50.
Mouse Anti-TSC2 Monoclonal Antibody – Cat# MBS249468

Immunofluorescence (IF) of anti-TSC2 antibody

Fluorescent image of HeLa cell stained with TSC2 Antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with TSC2 primary antibody (1:25, 1 h at 37°C). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-mouse antibody (green) was used (1:400, 50 min at 37°C). Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37°C). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min). TSC2 immunoreactivity is localized to Microtubules significantly.

Western Blot (WB) of anti-TSC2 antibody

TSC2 Antibody western blot of mouse NIH-3T3 cell line lysates (35 ug/lane). The TSC2 antibody detected the TSC2 protein (arrow).

NCBI Summary for TSC2

Mutations in this gene lead to tuberous sclerosis complex. Its gene product is believed to be a tumor suppressor and is able to stimulate specific GTPases. The protein associates with hamartin in a cytosolic complex, possibly acting as a chaperone for hamartin. Alternative splicing results in multiple transcript variants encoding different isoforms. UniProt Comments for TSC2

TSC2: a product of the tumor suppressor gene TSC2. Tuberin and Hamartin (TSC1) form a tumor suppressor heterodimer that inhibits the mTOR nutrient signaling input. TSC1/TSC2 targets the small G protein Rheb, a novel mediator of the nutrient signaling input to mTOR. Functions as a Rheb GTPase activating protein (GAP). Four alternatively spliced isoforms have been described.
