Application Notes for anti-CD14 antibody
PBMC: Add 10 ul of MAB/106 PBMC in 100 ul PBS. Mix gently and incubate for 15 minutes at 2 to 8°C. Wash twice with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. WHOLE BLOOD: Add 10 ul of MAB/100 ul of whole blood. Mix gently and incubate for 15 minutes at room temperature (20°C). Lyse the whole blood. Wash once with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. See instrument manufacturer's instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope. ALLOPHYCOCYANIN: (APC) conjugates are analyzed in multi-color flow cytometry with instruments equipped with a second laser and proper filters. Laser excitation is at 633 nm with a Helium Neon (HeNe) laser or a 600-640 nm (633 nm) range for a Dye laser. Peak fluorescence emission is at 660 nm.

Other Notes

Small volumes of anti-CD14 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CD14

The protein encoded by this gene is a surface antigen that is preferentially expressed on monocytes/macrophages. It cooperates with other proteins to mediate the innate immune response to bacterial lipopolysaccharide. Alternative splicing results in multiple transcript variants encoding the same protein. UniProt Comments for CD14

Function: In concert with LBP, binds to monomeric lipopolysaccharide and delivers it to the MD-2/TLR4 complex, thereby mediating the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Up-regulates cell surface molecules, including adhesion molecules. Ref.15

Subunit structure: Belongs to the lipopolysaccharide (LPS) receptor, a multi-protein complex containing at least CD14, MD-2 and TLR4.

Subcellular location: Cell membrane; Lipid-anchor › GPI-anchor.

Tissue specificity: Expressed strongly on the surface of monocytes and weakly on the surface of granulocytes; also expressed by most tissue macrophages.

Post-translational modification: N- and O- glycosylated. O-glycosylated with a core 1 or possibly core 8 glycan.

Sequence similarities: Contains 11 LRR (leucine-rich) repeats.
