Product Overview

Product Name
Pegylated leptin, Active Protein
Full Product Name
Pegylated rat leptin
Product Gene Name
Product Synonym Names
Rat pegylated leptin
Sequence
The sequence of the N-terminal amino acids of pegylated recombinan t rat leptin could not be determined indicating that the PEG moiety is attached to the N-terminus .
The sequence of the N-terminal amino acids could not be determined indicating that the PEG moiety is attached to the N-terminus .
Host
Escherichia Coli
Form/Format
White lyophilized (freeze-dried) powder.
Purity/Purification
The purity of pegylated recombinant rat leptin is greater than 99.0% as determined by:
(a) Gel filtration analysis.
(b) Analysis by reducing and non-reducing SDS-PAGE gel.
Preparation and Storage
Lyophilized pegylated recombinant rat leptin although stable at room temperature for several weeks, should be stored desiccated below -18 degree C. Upon reconstitution of pegylated recombinant rat leptin at > 0.1 mg/ml and up to 2 mg/ml and filter sterilization pegylated recombinant rat leptin can be stored at 4 degree C or even room temperature for several weeks making it suitable for long term infusion studies using osmotic pumps. At lower concentration of pegylated recombinant rat leptin addition of a carrier protein (0.1% HSA or BSA) is suggested. Please prevent freeze-thaw cycles.
Molecular Weight
18,641 Da
OMIM
164160
3D Structure
ModBase 3D Structure for P41159
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Solubility
It is recommended to reconstitute the lyophilized pegylated recombinant rat leptin in sterile water or sterile 0.4% NaHCO3 adjusted to pH 8-9, not less than 100ug/ml, which can then be further diluted with other aqueous solutions.
Protein Content
Protein quantitation of pegylated recombinant rat leptin was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.200 as the extinction coefficient for a 0.1% (1mg/ml) protein solution at pH 8.0. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
Dimers and Aggregates
The purified pegylated recombinant rat leptin consists of > 95% monomers as determined by gel-filtration chromatography.
Endotoxin
Less than 0.05 ng/ug (0.5 IEU/ug) of pegylated recombinant rat leptin.
Formulation
The pegylated recombinant rat leptin was lyophilized from a concentrated (0.65mg/ml) solution with 0.003mM NaHCO3 and the powder contains 35 to 40% protein.
Biological Activity
pegylated recombinant rat leptin is capable of stimulatng proliferation of BAF/3 cells stably transfected with the long form of human leptin receptor. Its in vitro activity is only slightly lower than the non-pegylated recombinant rat leptin but in vivo it has profound weight reducing effect (as compared to the non-pegylated leptin), resulting mainly from reduced food intake.

Other Notes

Small volumes of LEP active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for LEP

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. UniProt Comments for LEP

leptin: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Belongs to the leptin family.
