IGF-I (rbIGF-1) Rabbit Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS400111

IGF-1 is a small protein secreted mainly but not solely by the liver and circulating in blood mostly as a complex with various IGF binding proteins in the blood. It has growth-regulating, insulin-like, and mitogenic activities and it is secreted in response… More details

Product Overview

Online Datasheet in PDF
Product Name
IGF-I (rbIGF-1), Active Protein
Full Product Name
Rabbit IGF-I
Product Gene Name
Product Synonym Names
Rabbit insulin-like growth factor 1 (rbIGF-1)
Sequence
MTAGPETLCG AELVDALQFV CGDRGFYFNK PTGYGSSSRR APQTGIVDEC CFRSCDLRRL EMYCAPLKP
Host
E Coli
Form/Format
White lyophilized powder.
Purity/Purification
Greater than 98.0% as determined by:
(a) Analysis by reducing and non-reducing SDS-PAGE gel.
(b) Gel-filtration chromatography under non denaturing conditions.
Preparation and Storage
Lyophilized rbIGF-I although stable at room temperature for several weeks, should be stored desiccated below -18 degree C. Upon reconstitution at > 0.1 rbIGF-I mg/ml and up to 2 2 mg/ml and filter sterilization rbIGF-I can be stored at +4 degree C.
Molecular Weight
17,066 Da
3D Structure
ModBase 3D Structure for P07455
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Solubility
It is recommended to reconstitute the lyophilized rbIGF-I in sterile 0.4% NaHCO3 adjusted, not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Protein Content
Protein quantization was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.60 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the DNAman computer analysis program of protein sequences.
Dimers and Aggregates
The purified rbIGF-I consists of > 95% monomers as determined by gel-filtration chromatography.
Endotoxin
Endotoxin level is <0.5 EU/ug.
Formulation
The protein was lyophilized from a concentrated (0.5 mg/ml) solution with 0.02% NaHCO3.
Biological Activity
rbIGF-I is biologically active when compared to human IGF-1. The ED50, calculated by the dose -dependent proliferation of human MCF/7 cells is 5 to 25 ng/ml in the cell culture mixture dependent on culture conditions. Its activity consisits of 30-40 % compared to human IGF-1.

Other Notes

Small volumes of rbIGF-1 active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for rbIGF-1

The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. Stimulates glucose transport in bone-derived osteoblastic (PyMS) cells and is effective at much lower concentrations than insulin, not only regarding glycogen and DNA synthesis but also with regard to enhancing glucose uptake. May play a role in synapse maturation. Ca2+-dependent exocytosis of IGF1 is required for sensory perception of smell in the olfactory bulb. Acts as a ligand for IGF1R. Binds to the alpha subunit of IGF1R, leading to the activation of the intrinsic tyrosine kinase activity which autophosphorylates tyrosine residues in the beta subunit thus initiatiating a cascade of down-stream signaling events leading to activation of the PI3K-AKT/PKB and the Ras-MAPK pathways. Binds to integrins ITGAV:ITGB3 and ITGA6:ITGB4. Its binding to integrins and subsequent ternary complex formation with integrins and IGFR1 are essential for IGF1 signaling. Induces the phosphorylation and activation of IGFR1, MAPK3/ERK1, MAPK1/ERK2 and AKT1 ().
