The 14.3.3 family of proteins were originally discovered as prominent protein spots on 2 dimensional gels. They are a family of 28-33kDa proteins which are often major proteins of the cytoplasm (1). They function as binding partners for phosphoserine and phosphthreonine sites in other proteins, though they have other binding partners which are not dependent on phosphorylation. These binding interactions are important in the regulation of molecules such as the signaling kinases in the MAP kinase pathway c-Raf and b-Raf, the proapoptotic molecules Bad and Bax and the cell cycle regulator Cdc25. There are 7 mammalian 14.3.3 proteins and they are normally expressed as homo or in some cases heterodimers. 14.3.3η or 14.3.3 eta is widely expressed and concentrated in the nervous system. The HGNC name for 14.3.3 is YWHAH, and an alternate name is tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein 1, obviously due to a role for this protein in activation of tyrosine and tryptophan hydroxylases. One result of this is that the HGNC name is YWHAH. The 14.3.3η protein accumulates in the CSF of patients suffering form Creutzfeld Jacob Disease, and can be used for the this disease (2). Furthermore, this protein binds alphasynuclein in the Lewy bodies of Parkinson's disease and has been linked to early-onset schizopherenia (3,4). The HGNC name for this protein is YWHAH.

NCBI Summary for 14-3-3-eta

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 99% identical to the mouse, rat and bovine orthologs. This gene contains a 7 bp repeat sequence in its 5' UTR, and changes in the number of this repeat have been associated with early-onset schizophrenia and psychotic bipolar disorder. UniProt Comments for 14-3-3-eta

Function: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. Negatively regulates the kinase activity of PDPK1. Ref.13

Subunit structure: Homodimer

By similarity. Interacts with many nuclear hormone receptors and cofactors including AR, ESR1, ESR2, MC2R, NR3C1, NRIP1, PPARBP and THRA. Interacts with ABL1 (phosphorylated form); the interaction retains it in the cytoplasm. Interacts with ARHGEF28 and CDK16

By similarity. Weakly interacts with CDKN1B. Interacts with GAB2. Interacts with KCNK18 in a phosphorylation-dependent manner. Interacts with SAMSN1

By similarity. Interacts with the 'Ser-241' phosphorylated form of PDPK1. Ref.12 Ref.13 Ref.14 Ref.15 Ref.16 Ref.19

Tissue specificity: Expressed mainly in the brain and present in other tissues albeit at lower levels.

Post-translational modification: Phosphorylated on Ser-59 by protein kinase C delta type catalytic subunit in a sphingosine-dependent fashion

By similarity.

Sequence similarities: Belongs to the 14-3-3 family.
