Application Notes for anti-SETD2 antibody
Peptide ELISA: Antibody detection limit dilution 1: 128000.
Western Blot: Approx 290kDa band observed in lysates of mouse fibroblasts. The observed band is knocked down by two different siRNA molecules. Calculated MW of 231kDa according to NP_054878.3). Data obtained from anonymous customer and obtained from a previous batch (from a different goat). The Ponceau red demonstrates proper protein loading. Recommended concentration: 0.5-2ug/ml.

Western Blot (WB) of anti-SETD2 antibody

(0.5ug/ml) staining of mouse fibroblast lysates transiently expressing different siRNA in second and third lane (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Other Notes

Small volumes of anti-SETD2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for SETD2

Huntington's disease (HD), a neurodegenerative disorder characterized by loss of striatal neurons, is caused by an expansion of a polyglutamine tract in the HD protein huntingtin. This gene encodes a protein belonging to a class of huntingtin interacting proteins characterized by WW motifs. This protein is a histone methyltransferase that is specific for lysine-36 of histone H3, and methylation of this residue is associated with active chromatin. This protein also contains a novel transcriptional activation domain and has been found associated with hyperphosphorylated RNA polymerase II. [provided by RefSeq, Aug 2008]

UniProt Comments for SETD2

SETD2: Histone methyltransferase that methylates 'Lys-36' of histone H3. H3 'Lys-36' methylation represents a specific tag for epigenetic transcriptional activation. Probably plays a role in chromatin structure modulation during elongation via its interaction with hyperphosphorylated POLR2A. Binds DNA at promoters. May also act as a transcription activator that binds to promoters. Binds to the promoters of adenovirus 12 E1A gene in case of infection, possibly leading to regulate its expression. Belongs to the histone-lysine methyltransferase family. SET2 subfamily. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 2.1.1.43; Methyltransferase; Methyltransferase, protein lysine; Amino Acid Metabolism - lysine degradation

Chromosomal Location of Human Ortholog: 3p21.31

Cellular Component: chromosome; nucleoplasm

Molecular Function: histone lysine N-methyltransferase activity (H3-K36 specific); histone-lysine N-methyltransferase activity; protein binding

Biological Process: angiogenesis; embryonic cranial skeleton morphogenesis; forebrain development; mesoderm morphogenesis; mismatch repair; morphogenesis of a branching structure; neural tube closure; pericardium development; regulation of transcription, DNA-dependent; RNA elongation from RNA polymerase II promoter; stem cell development
