Application Notes for anti-HMGA1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 32000.
Immunohistochemistry: In paraffin embedded Human Placenta shows strong nuclear staining. Recommended concentration, 4-6ug/ml.
Western Blot: Preliminary experiments gave an approx 50kDa band in Human Testis and Ovary lysates after 0.3ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 11.7kDa according to isoform a (also called HMG-I; NP_665906.1; NP_665908.1; NP_665911.1) and 10.7kDa according to isoform b((also called HMG-Y; NP_002122.1; NP_665909.1; NP_665910.1; NP_665912.1). The 50kDa band was successfully blocked by incubation with the immunizing peptide.

Immunohistochemistry (IHC) of anti-HMGA1 antibody

(4ug/ml) staining of paraffin embedded Human Placenta. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

Other Notes

Small volumes of anti-HMGA1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for HMGA1

This gene encodes a chromatin-associated protein involved in the regulation of gene transcription, integration of retroviruses into chromosomes, and the metastatic progression of cancer cells. The encoded protein preferentially binds to the minor groove of AT-rich regions in double-stranded DNA. Multiple transcript variants encoding different isoforms have been found for this gene. Pseudogenes of this gene have been identified on multiple chromosomes. [provided by RefSeq, Jan 2016]

UniProt Comments for HMGA1

HMGA1: HMG-I/Y bind preferentially to the minor groove of A+T rich regions in double stranded DNA. It is suggested that these proteins could function in nucleosome phasing and in the 3'-end processing of mRNA transcripts. They are also involved in the transcription regulation of genes containing, or in close proximity to A+T-rich regions. A chromosomal aberration involving HMGA1 is found in pulmonary chondroid hamartoma. Translocation t(6;14)(p21;q23-24) with RAD51B. Belongs to the HMGA family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: DNA-binding

Chromosomal Location of Human Ortholog: 6p21

Cellular Component: cytosol; focal adhesion; nucleoplasm; nucleus; transcription factor complex

Molecular Function: 5'-deoxyribose-5-phosphate lyase activity; AT DNA binding; chromatin binding; DNA binding; DNA-(apurinic or apyrimidinic site) lyase activity; enzyme binding; ligand-dependent nuclear receptor transcription coactivator activity; peroxisome proliferator activated receptor binding; protein binding; retinoic acid receptor binding; retinoid X receptor binding; transcription factor activity; transcription factor binding
