Application Notes for anti-M6PR antibody
Peptide ELISA: Antibody detection limit dilution 1: 4000.
Western Blot: Preliminary experiments gave bands at approx 70kDa and 35kDa in Human Lymph Node and Thyroid Gland lysates after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 31. 0kDa according to NP_002346.1. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).

Other Notes

Small volumes of anti-M6PR antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for M6PR

This gene encodes a member of the P-type lectin family. P-type lectins play a critical role in lysosome function through the specific transport of mannose-6-phosphate-containing acid hydrolases from the Golgi complex to lysosomes. The encoded protein functions as a homodimer and requires divalent cations for ligand binding. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. A pseudogene of this gene is located on the long arm of chromosome X. [provided by RefSeq, May 2011]

UniProt Comments for M6PR

M6PR: Transport of phosphorylated lysosomal enzymes from the Golgi complex and the cell surface to lysosomes. Lysosomal enzymes bearing phosphomannosyl residues bind specifically to mannose-6- phosphate receptors in the Golgi apparatus and the resulting receptor-ligand complex is transported to an acidic prelyosomal compartment where the low pH mediates the dissociation of the complex.

Protein type: Membrane protein, integral

Chromosomal Location of Human Ortholog: 12p13

Cellular Component: endosome; integral to plasma membrane; late endosome; lysosomal membrane; membrane; perinuclear region of cytoplasm; trans-Golgi network; trans-Golgi network membrane; transport vesicle

Molecular Function: mannose binding; mannose transmembrane transporter activity; transmembrane receptor activity

Biological Process: endosome to lysosome transport; mannose transport; receptor-mediated endocytosis; signal transduction
