Application Notes for anti-FOXI1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 32000.
Immunohistochemistry: In paraffin embedded Human Testis shows primarily nuclear staining in the primary spermatocytes. Recommended concentration, 2-4ug/ml. This product has been successfully used in Mouse Epididymus by anonymous customer.
Western Blot: Preliminary experiments gave bands at approx 50 kDa and 25 kDa in Human Kidney lysate after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the predicted sizes of 41. 0kDa according to NP_036320 (isoform a) and 30.8kDa according to NP_65892 (isofrom b). The bands were successfully blocked by incubation with the immunizing peptide.

Immunohistochemistry (IHC) of anti-FOXI1 antibody

(2ug/ml) staining of paraffin embedded Human Testis. Steamed antigen retrieval with citrate buffer pH 6, HRP-staining.

Other Notes

Small volumes of anti-FOXI1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for FOXI1

This gene belongs to the forkhead family of transcription factors which is characterized by a distinct forkhead domain. The specific function of this gene has not yet been determined; however, it is possible that this gene plays an important role in the development of the cochlea and vestibulum, as well as embryogenesis. Mutations in this gene may be associated with the common cavity phenotype. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for FOXI1

FOXI1: Transcriptional activator required for the development of normal hearing, sense of balance and kidney function. Required for the expression of SLC26A4/PDS, JAG1 and COCH in a subset of epithelial cells and the development of the endolymphatic system in the inner ear. Also required for the expression of SLC4A1/AE1, SLC4A9/AE4, ATP6V1B1 and the differentiation of intercalated cells in the epithelium of distal renal tubules. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: DNA-binding

Chromosomal Location of Human Ortholog: 5q34

Cellular Component: nucleus

Molecular Function: DNA bending activity; sequence-specific DNA binding; transcription factor activity

Biological Process: embryonic development; inner ear morphogenesis; positive regulation of transcription from RNA polymerase II promoter; transcription from RNA polymerase II promoter

Disease: Deafness, Autosomal Recessive 4, With Enlarged Vestibular Aqueduct; Pendred Syndrome
