Application Notes for anti-FCGR2B antibody
Peptide ELISA: Antibody detection limit dilution 1: 16000.
Immunohistochemistry: In paraffin embedded Human Liver shows membranous staining in endothelial cells between the hepatocytes. Similar results were obtained in Human Placenta. Recommended concentration, 3-6ug/ml.
Western Blot: Approx 34kDa band observed in Human myeloid erythroblast cell line K562 and Human monocytic cell line U937 lysates (calculated MW of 34. 0kDa according to NP_003992.3;). In transfected HEK293 transiently expressing CD32 a band of approx. 40kDa (glycoprotein) is observed. This band is not observed in the non-transfected HEK293. Recommended concentration: 0.3-1ug/ml.

Western Blot (WB) of anti-FCGR2B antibody

(0.3ug/ml) staining of K562 lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.
Goat Anti-FCGR2B Polyclonal Antibody – Cat# MBS420954

Western Blot (WB) of anti-FCGR2B antibody

HEK293 overexpressing CD32 (RC211982) and probed (mock transfection in first lane), tested by Origene.
Goat Anti-FCGR2B Polyclonal Antibody, Western Blot (WB) – Cat# MBS420954

Immunohistochemistry (IHC) of anti-FCGR2B antibody

(3.8ug/ml) staining of paraffin embedded Human Liver. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.
Goat Anti-FCGR2B Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS420954

Other Notes

Small volumes of anti-FCGR2B antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for FCGR2B

The protein encoded by this gene is a low affinity receptor for the Fc region of immunoglobulin gamma complexes. The encoded protein is involved in the phagocytosis of immune complexes and in the regulation of antibody production by B-cells. Variations in this gene may increase susceptibilty to systemic lupus erythematosus (SLE). Several transcript variants encoding different isoforms have been found for this gene. UniProt Comments for FCGR2B

FCGR2B: a transmembrane receptor for the Fc region of complexed or aggregated immunoglobulin gamma (IgG). Involved in a variety of effector and regulatory functions such as phagocytosis of immune complexes and modulation of antibody production by B-cells. Binding to this receptor results in down modulation of previous state of cell activation triggered via antigen receptors on B cells, T cells or via another Fc receptor. Three splice-variant isoforms have been observed.

Protein type: Cell surface; Membrane protein, integral; Oncoprotein
