Application Notes for anti-SECISBP2 antibody
Peptide ELISA: Antibody detection limit dilution 1: 64000.
Immunohistochemistry: In paraffin embedded Human Testis shows granular cytoplasm staining in Leydig cells. Recommended concentration, 3ug/ml.
Western Blot: Approx 120kDa band detected in HFF cell lysates. Data kindly provided by Laura Papp and Kum Kum Khanna. The observed molecular weight corresponds to earlier findings in literature with different antibodies (Papp et al, Mol Cell Biol.2006 Jul;26(13): 4895-910; PMID: 16782878). Recommended concentration: 0.1-0.3ug/ml.
Immunocytochemsitry/Immunofluorescence: This antibody has been successfully used in ICC/IF for C33A cells as described in the following paper: Papp et al, Nucleic Acids Res.2008 Dec;36(22): 7192-206, PMID: 19004874.
Immunoprecipitation: This antibody has also been successfully used for IP.

Western Blot (WB) of anti-SECISBP2 antibody

Staining of HFF cell lysate with B) 100nM siRNA or A) control siRNA. Detected by chemiluminescence. Data kindly provided by Laura Papp and Kum Kum Khanna, Brisbane, Australia.

NCBI Summary for SECISBP2

The incorporation of selenocysteine into a protein requires the concerted action of an mRNA element called a sec insertion sequence (SECIS), a selenocysteine-specific translation elongation factor and a SECIS binding protein. With these elements in place, a UGA codon can be decoded as selenocysteine. The gene described in this record encodes a nuclear protein that functions as a SECIS binding protein. Mutations in this gene have been associated with a reduction in activity of a specific thyroxine deiodinase, a selenocysteine-containing enzyme, and abnormal thyroid hormone metabolism. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Sep 2013]

UniProt Comments for SECISBP2

SECISBP2: Binds to the SECIS element in the 3'-UTR of some mRNAs encoding selenoproteins. Binding is stimulated by SELB. Defects in SECISBP2 are a cause of abnormal thyroid hormone metabolism (ATHYHM). This phenotype is associated with a reduction in type II iodothyronine deiodinase activity. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Translation

Chromosomal Location of Human Ortholog: 9q22.2

Cellular Component: mitochondrion; nucleoplasm; ribonucleoprotein complex

Molecular Function: mRNA 3'-UTR binding; protein binding; ribonucleoprotein binding

Biological Process: neuron development; selenocysteine incorporation; striatum development

Disease: Thyroid Hormone Metabolism, Abnormal
