Peptide ELISA: Antibody detection limit dilution 1: 128000.
Western Blot: Approx 33kDa band observed in human Heart lysate (calculated MW of 32.1kDa according to NP_003653.1). In transfected HEK293 transiently expressing PIR a band of approx.35kDa is observed. This band is not observed in the non-transfected HEK293. Recommended concentration: 0.3 - 1. 0 ug/ml.

Western Blot (WB) of anti-PIR antibody

(0.3ug/ml) staining of Human Heart lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.
Goat Anti-PIR Polyclonal Antibody – Cat# MBS421077

Western Blot (WB) of anti-PIR antibody

HEK293 overexpressing PIR (RC223111) and probed (mock transfection in first lane), tested by Origene.
Goat Anti-PIR Polyclonal Antibody, Western Blot (WB) – Cat# MBS421077

Other Notes

Small volumes of anti-PIR antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PIR

This gene encodes a member of the cupin superfamily. The encoded protein is an Fe(II)-containing nuclear protein expressed in all tissues of the body and concentrated within dot-like subnuclear structures. Interactions with nuclear factor I/CCAAT box transcription factor as well as B cell lymphoma 3-encoded oncoprotein suggest the encoded protein may act as a transcriptional cofactor and be involved in the regulation of DNA transcription and replication. Alternatively spliced transcript variants have been described. [provided by RefSeq, Jul 2008]

UniProt Comments for PIR

PIR: Possible transcriptional coregulator. May contribute to the regulation of cellular processes via its interaction with BCL3. May be required for efficient terminal myeloid maturation of hematopoietic cells. May play a role in the regulation of cell migration. May promote apoptosis when overexpressed. Has quercetin 2,3-dioxygenase activity (in vitro). May interact with NF1/CTF1. Interacts with BCL3. Identified in a complex comprised of PIR, BLC3, NFKB1 and target DNA. Up-regulated in CD34(+) cells upon myelomonocytic differentiation. Down-regulated in many acute myeloid leukemias. Up-regulated in primary bronchial epithelial cells exposed to cigarette smoke extract. Highly expressed in a subset of melanomas. Detected at very low levels in most tissues. Expressed in all tissues, with highest level of expression in heart and liver. Inhibited by kojic acid, sodium diethyldithiocarbamate and 1,10-phenanthroline monohydrochloride. Belongs to the pirin family.

Protein type: EC 1.13.11.24

Chromosomal Location of Human Ortholog: Xp22.2

Cellular Component: cytoplasm; nucleus

Molecular Function: metal ion binding; protein binding; quercetin 2,3-dioxygenase activity; transcription cofactor activity

Biological Process: monocyte differentiation; regulation of transcription, DNA-dependent; transcription from RNA polymerase II promoter
