Peptide ELISA: Antibody detection limit dilution 1: 64000.
Western Blot: Approx 30kDa band observed in human kidney and human placenta lysates (calculated MW of 28.8kDa according to NP_039234.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Edwards et al, Am J Physiol.1999 Mar;276(3 Pt 2): F398-408.; PMID: 10070163). Recommended concentration: 0.1-0.3ug/ml.
Immunofluorescence: Min6 cells transiently expressing mouse Clic4 shows cytoplasmic staining of two cells. Recommended concentration, 5ug/ml with overnight incubation.

Western Blot (WB) of anti-CLIC4 antibody

(0.1ug/ml) staining of human kidney lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Other Notes

Small volumes of anti-CLIC4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CLIC4

Chloride channels are a diverse group of proteins that regulate fundamental cellular processes including stabilization of cell membrane potential, transepithelial transport, maintenance of intracellular pH, and regulation of cell volume. Chloride intracellular channel 4 (CLIC4) protein, encoded by the CLIC4 gene, is a member of the p64 family; the gene is expressed in many tissues and exhibits a intracellular vesicular pattern in Panc-1 cells (pancreatic cancer cells). UniProt Comments for CLIC4

CLIC4: Can insert into membranes and form poorly selective ion channels that may also transport chloride ions. Channel activity depends on the pH. Membrane insertion seems to be redox-regulated and may occur only under oxydizing conditions. Promotes cell- surface expression of HRH3. Has alternate cellular functions like a potential role in angiogenesis or in maintaining apical- basolateral membrane polarity during mitosis and cytokinesis. Could also promote endothelial cell proliferation and regulate endothelial morphogenesis (tubulogenesis). Belongs to the chloride channel CLIC family.

Protein type: Membrane protein, integral

Chromosomal Location of Human Ortholog: 1p36.11

Cellular Component: actin cytoskeleton; apical part of cell; cell surface; centrosome; cytoplasm; cytoplasmic vesicle membrane; cytosol; intercellular junction; intracellular; microtubule cytoskeleton; microvillus; midbody; mitochondrion; nuclear matrix; perinuclear region of cytoplasm; plasma membrane

Molecular Function: chloride channel activity; glutathione transferase activity; protein binding; voltage-gated ion channel activity

Biological Process: angiogenesis; apoptosis; branching morphogenesis of a tube; cell differentiation; chloride transport; endothelial cell morphogenesis; establishment and/or maintenance of apical/basal cell polarity; fertilization; glutathione metabolic process; keratinocyte differentiation; multicellular organism growth; negative regulation of cell migration; regulation of cytoskeleton organization and biogenesis; vacuolar acidification
