Application Notes for anti-CES1 antibody
Peptide ELISA: Antibody detection limit dilution 1: 8000.
Immunohistochemistry: In paraffin embedded Human Lung shows staining of lymphocytes in the vasculature. Recommended concentration, 2-3ug/ml.
Western Blot: Approx 55-60kDa band observed in Human Liver and Ileum lysates (calculated MW of 62.6kDa according to NP_001020366.1). In transfected HEK293 transiently expressing CES1 a band of approx. 65kDa is observed. This band is not observed in the non-transfected HEK293. Recommended concentration: 0. 03-0.1ug/ml.

Western Blot (WB) of anti-CES1 antibody

(0. 03ug/ml) staining of Human Liver lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Immunohistochemistry (IHC) of anti-CES1 antibody

(2.5ug/ml) staining of paraffin embedded Human Lung. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

Other Notes

Small volumes of anti-CES1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CES1

This gene encodes a member of the carboxylesterase large family. The family members are responsible for the hydrolysis or transesterification of various xenobiotics, such as cocaine and heroin, and endogenous substrates with ester, thioester, or amide bonds. They may participate in fatty acyl and cholesterol ester metabolism, and may play a role in the blood-brain barrier system. This enzyme is the major liver enzyme and functions in liver drug clearance. Mutations of this gene cause carboxylesterase 1 deficiency. Three transcript variants encoding three different isoforms have been found for this gene. [provided by RefSeq, Jun 2010]

UniProt Comments for CES1

CES1: Involved in the detoxification of xenobiotics and in the activation of ester and amide prodrugs. Hydrolyzes aromatic and aliphatic esters, but has no catalytic activity toward amides or a fatty acyl-CoA ester. Hydrolyzes the methyl ester group of cocaine to form benzoylecgonine. Catalyzes the transesterification of cocaine to form cocaethylene. Displays fatty acid ethyl ester synthase activity, catalyzing the ethyl esterification of oleic acid to ethyloleate. Belongs to the type-B carboxylesterase/lipase family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 3.1.1.1; EC 3.1.1.56; Hydrolase; Xenobiotic Metabolism - drug metabolism - other enzymes

Chromosomal Location of Human Ortholog: 16q22.2

Cellular Component: endoplasmic reticulum lumen; extracellular space

Molecular Function: methylumbelliferyl-acetate deacetylase activity
