Application Notes for anti-TNKS2 antibody
Western Blot: In transfected HEK293 transiently expressing TANK2 a band of approx.127kDa is observed. This band is not observed in the non-transfected HEK293. The calculated molecular size is 127 kDa according to NP_079511.1. Recommended concentration, 0.5-1.5ug/ml.

Western Blot (WB) of anti-TNKS2 antibody

HEK293 overexpressing TANK2 (lane 4) and TANK1 (lane 2) and probed (mock transfection in first lane). Lane three is empty. Lower panel shows the same lysates probed for alpha-Actin to show protein levels. Primary incubation (0.5ug/ml) was overnight at 4 degree C. Detected by chemiluminescence.

Other Notes

Small volumes of anti-TNKS2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for TNKS2

TNKS2: Poly-ADP-ribosyltransferase involved in various processes such as Wnt signaling pathway, telomere length and vesicle trafficking. Acts as an activator of the Wnt signaling pathway by mediating poly-ADP-ribosylation of AXIN1 and AXIN2, 2 key components of the beta-catenin destruction complex: poly-ADP- ribosylated target proteins are recognized by RNF146, which mediates their ubiquitination and subsequent degradation. Also mediates poly-ADP-ribosylation of BLZF1 and CASC3, followed by recruitment of RNF146 and subsequent ubiquitination. Mediates poly-ADP-ribosylation of TERF1, thereby contributing to the regulation of telomere length. May also regulate vesicle trafficking and modulate the subcellular distribution of SLC2A4/GLUT4-vesicles.

Protein type: Nuclear envelope; Transferase; EC 2.4.2.30

Chromosomal Location of Human Ortholog: 10q23.3

Cellular Component: cytoplasm; cytosol; Golgi membrane; nuclear chromosome, telomeric region; nuclear envelope; nucleus; pericentriolar material; perinuclear region of cytoplasm

Molecular Function: enzyme binding; metal ion binding; NAD+ ADP-ribosyltransferase activity; protein binding

Biological Process: multicellular organism growth; positive regulation of telomere maintenance via telomerase; protein amino acid ADP-ribosylation; protein polyubiquitination; regulation of multicellular organism growth; Wnt receptor signaling pathway
