Application Notes for anti-DTX3 antibody
Peptide ELISA: Antibody detection limit dilution 1: 8000.
Western Blot: Approx 90kDa band observed in Mouse Fetal Brain lysates and in lysates of cell line NIH3T3 (calculated MW of 38. 0kDa according to Human NP_848597.1 and 37.98kDa to Mouse NP_109639.1). The observed much higher molecular weight corresponds to earlier findings in literature with different antibodies (Takeyama et al, J Biol Chem.2003 Jun13;278(24): 21930-7.; PMID: 12670957). Recommended concentration: 0.2-0.5ug/ml.

Western Blot (WB) of anti-DTX3 antibody

(0.3ug/ml) staining of NIH3T3 lysate (35ug protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.

Other Notes

Small volumes of anti-DTX3 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for DTX3

DTX3 functions as an E3 ubiquitin ligase (Takeyama et al., 2003 [PubMed 12670957]).[supplied by OMIM, Nov 2009]

UniProt Comments for DTX3

DTX3: Regulator of Notch signaling, a signaling pathway involved in cell-cell communications that regulates a broad spectrum of cell-fate determinations. Probably acts both as a positive and negative regulator of Notch, depending on the developmental and cell context. Functions as an ubiquitin ligase protein in vitro, suggesting that it may regulate the Notch pathway via some ubiquitin ligase activity. Belongs to the Deltex family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Ubiquitin conjugating system; EC 6.3.2.-; Ubiquitin ligase; Ligase

Chromosomal Location of Human Ortholog: 12q13.3

Cellular Component: cytoplasm

Molecular Function: ligase activity; protein binding; zinc ion binding

Biological Process: Notch signaling pathway; protein ubiquitination
